You could make your 1:1000 dilution in a lot of ways.

You could do 1:10, then 1:10, then 1:10.

or 1:50, then 1:20

or 1:2, then 1:500

or 1:4, then 1:250

or 1:25, then 1:40

What might you think about when you are choosing?

THE BOSS has some suggestions.

"Don't make up a lot of the solution. I don't want to see any dilutions larger than 1:100! If you make more than we need, we have to pay for disposing of what you don't use -- and you'll have trouble mixing a large volume properly, anyway."

"But don't be too stingy. You can't save on cell cultures by taking just a tiny sample of the stock culture - cells aren't that evenly distributed, and a sample smaller than 0.1 mL won't have the same concentration as the whole culture."

"Do the smallest dilution first. If you cut something 1:2, it's still a pretty concentrated stock solution. We can use your leftovers to make other solutions around the lab. If you cut it 1:100 right off the bat, it's no use to anyone else."

"If you're making a dilution of 1:3400, or something of that sort, do the 1:34 dilution first. That's the trickiest measuring; you should do it to make sure it's done right. Then if we have to make up some more working solution later, we can take your leftover and just cut it 1:100. Any idiot can do that!"
